What is Melanotan 1?
Melanotan 1, known generically as afamelanotide, is a synthetic analog of alpha-melanocyte-stimulating hormone, the natural signal that tells pigment cells in the skin to produce melanin. It is the same active ingredient as Scenesse, an FDA-approved implant for a rare light-sensitivity disorder. Melanotan 2 is a different peptide with a very different safety record. Here melanotan 1 is prescribed as a take-home vial after a free consultation and a review of your history.
Melanotan 1 benefits and uses.
-
Deeper skin pigment
Darkening of the skin is a documented effect of afamelanotide in its human trials, the same receptor action that protects against light.
-
Sun tolerance
In two randomized, placebo-controlled trials, afamelanotide increased pain-free time in sunlight for adults with erythropoietic protoporphyria.
-
Vitiligo repigmentation
A small human study combined afamelanotide with light therapy for vitiligo.
- 168adults in the two trials behind its FDA approval
- 2019the year the FDA approved Scenesse, the same active ingredient
- Every vialcomes with its own certificate of analysis
How does Melanotan 1 work?
Alpha-MSH acts on a receptor on pigment cells called MC1R. When it binds, the cells produce more eumelanin, the brown-black pigment that absorbs light and protects the skin. Afamelanotide binds the same receptor more strongly and for longer than the natural signal, which is why it darkens skin and, in its approved use, lets people tolerate more light before pain begins.
What does the research on Melanotan 1 show?
- Human study
Two randomized, placebo-controlled trials in 168 adults with erythropoietic protoporphyria, published in the New England Journal of Medicine, supported its approval. Langendonk 2015, NEJM
- Human study
An eight-year observational follow-up continued to track its use in that condition. Biolcati 2015
- Human study
A small human study combined afamelanotide with light therapy for vitiligo. Lim 2015

An independent 2023 systematic review summarized the afamelanotide evidence. Heerfordt 2023
Melanotan 1 side effects and safety.
- Nausea
- Headache
- Darkening of skin and moles, which is why regular skin checks are advised
- Melanotan 2 is a different peptide: FDA has flagged published reports of melanoma and other serious events with it.
These are the common effects on the approved product's label, which advises regular full-body skin checks because moles can darken. Tell the office about any mole that changes.

Common questions.
What is melanotan 1?
Melanotan 1, or afamelanotide, is a synthetic version of the hormone that tells pigment cells to make melanin. It is the same active ingredient as Scenesse, an FDA-approved implant.
Melanotan 1 vs melanotan 2: what is the difference?
They are different peptides. Melanotan 1 is afamelanotide, the active ingredient in an FDA-approved product. Melanotan 2 is a shorter peptide sold illicitly for tanning, and FDA has flagged published reports of melanoma, brain swelling, and priapism with it. Only melanotan 1 is prescribed here.
What does melanotan 1 do?
It increases eumelanin, the protective brown-black pigment in the skin. In human trials it increased pain-free time in sunlight for people with a rare light-sensitivity disorder, and skin darkening is a documented effect.
Is melanotan 1 safe?
On the approved product's label, the common effects are nausea, headache, and darkening of skin and moles, which is why regular skin checks are advised.
Is melanotan 1 FDA approved?
Its active ingredient is, as Scenesse, approved in 2019 for adults with erythropoietic protoporphyria. The take-home vial prescribed here is not the Scenesse implant.
How is melanotan 1 taken?
It is a small injection under the skin. Your dose and schedule are set by Catherine Seidle, APRN at your visit.
How much does melanotan 1 cost?
A take-home vial is $170, and it includes the bacteriostatic water and syringes. The consultation is free.
Does melanotan 1 replace sunscreen?
No. Keep using sun protection. Pigment adds some light tolerance, and it does not prevent sun damage on its own.
